目的建立测定兔眼球组织中高三尖杉酯碱(HHT)的含量及其分布。方法以三尖杉酯碱(HT)为内标 ,荧光法检测 ;眼球旁注射HHT ,分离各眼球组织分别制匀浆、萃取后进行测定。结果HHT与HT的保留时间 (tR)分别为6.35min和4.70min ;在5~200μg·L-1 线性相关系数为0.994。平均加样回收率为 (93.17±4.31) %。眼球旁注射HHT2mg 后兔眼各组织含量按巩膜、角膜、视网膜、玻璃体、晶状体依次下降 ,并且巩膜组织内含量远高于其他组织 ,但消除速率却呈相反顺序。结论球旁注射HHT可以分布至眼球各组织
Objective To establish a method for the determination of the content and distribution of homoharringtonine (HHT) in rabbit ocular tissues. Methods Hirschsilide (HT) was used as an internal standard and detected by fluorescence method. HHT was injected into the eyeball, and the eyeballs were isolated and homogenized separately for determination. Results The retention time (tR) of HHT and HT were 6.35min and 4.70min, respectively. The linear correlation coefficient between 5 and 200μg · L-1 was 0.994. The average recovery was (93.17 ± 4.31)%. The contents of sclera, cornea, retina, vitreous body and lens in the eye of rabbits after HHT2mg injection of eyeball decreased in turn, and the content of scleral tissue was much higher than that of other tissues, but the elimination rate was in reverse order. Conclusions The lateral injection of HHT can be distributed to all tissues of the eye
