目的:探讨生姜汁和干姜汁炮制黄连对乙醇致大鼠胃黏膜损伤的保护作用差异及其作用机制。方法:将大鼠随机分为7组,即空白组、模型组、生姜汁组、干姜汁组、生黄连组、生姜制黄连组、干姜制黄连组。采用乙醇致大鼠胃黏膜损伤,观察不同组大鼠胃黏膜损伤程度并计算胃黏膜损伤指数,取胃组织制成病理切片,光镜下观察大鼠胃黏膜组织学改变,采用酶联免疫吸附测定法测定血浆中白细胞介素-8(IL-8),肿瘤坏死因子-α(TNF-α)和6-酮-前列腺素F1α(6-keto-PGF_(1α))的含量。结果:与模型组比较,生姜汁组、干姜汁组、生黄连组、生姜制黄连组、干姜制黄连组大鼠胃黏膜损伤指数均显著下降(P<0.01)。与生黄连组比较,生姜制黄连组和干姜制黄连组大鼠胃黏膜损伤指数明显降低(P<0.05),且生姜制黄连组损伤指数较干姜制黄连组更低。血浆中IL-8和TNF-α的含量模型组与空白组相比显著上升(P<0.05),6-keto-PGF_(1α)的含量则显著下降(P<0.05),给药后IL-8和TNF-α含量均下降,6-keto-PGF_(1α)的含量则有所上升,且姜制后作用更强。结论:黄连经过姜汁炮制后可增强其保护胃黏膜损伤的作用,作用机制可能是与抑制IL-8,TNF-α的产生和促进6-keto-PGF_(1α)的产生有关,生姜汁的炮制效果较干姜汁好。
Objective: To investigate the protective effects of Coptis chinensis Franch and dried ginger juice on the gastric mucosal injury induced by ethanol in rats and its mechanism. Methods: The rats were randomly divided into 7 groups: blank group, model group, ginger juice group, dried ginger juice group, raw berberine group, ginger berberine group and rhizoma coptidis group. Gastric mucosal injury induced by ethanol was observed in rats. Gastric mucosal lesion was evaluated in different groups and gastric mucosal damage index was calculated. Gastric tissues were used to make pathological sections. Gastric mucosa histological changes were observed under light microscope. Enzyme-linked immunosorbent assay The levels of interleukin-8, tumor necrosis factor-α (TNF-α) and 6-keto-prostaglandin F1α (6-keto-PGF_ (1α) Results: Compared with the model group, the gastric mucosal injury index of the ginger juice group, the ginger juice group, the berberine group, the rhizoma coptidis group and the rhizome coptidis group were significantly decreased (P <0.01). Compared with the raw Coptidis group, the gastric mucosa injury index of the ginger group and the Coptis chinensis group were significantly decreased (P <0.05), and the damage index of the Coptis chinensis group was lower than those of the Coptis chinensis group. The contents of IL-8 and TNF-α in plasma in model group increased significantly (P <0.05) and the content of 6-keto-PGF 1α (P <0.05) 8 and TNF-α decreased, the content of 6-keto-PGF_ (1α) increased and the effect of ginger was stronger. CONCLUSION: Coptis chinensis Franch can enhance its protective effect on gastric mucosal injury after being prepared by ginger juice. The mechanism may be related to inhibiting the production of IL-8 and TNF-α and promoting the production of 6-keto-PGF_ (1α) Processing is better than ginger juice.
