目的探讨环氧化酶-2(COX-2)和细胞周期调节蛋白(Ki-67、cyclin A、p27)在食管鳞癌(ESCC)中的表达及其相关性。方法随机选取ESCC根治术后的切除标本102例和28例癌旁正常鳞状上皮组织。应用免疫组化检测COX-2、Ki-67、细胞周期蛋白A(cyclin A)和p27的表达。结果COX-2、Ki-67和cyclin A在ESCC表达明显高于癌旁正常鳞状上皮组织(P<0.01),p27在ESCC中表达明显低于癌旁正常鳞状上皮组织(P<0.01)。COX-2的表达与肿瘤的T分期有关,T1~T2期表达低于T3~T4期(P<0.05);Ki-67、cyclin A、p27的表达与细胞分化相关(P<0.01);COX-2与Ki-67、cyclin A表达呈正相关,与p27表达负相关。结论COX-2的表达程度可反映ESCC的肿瘤细胞浸润深度;COX-2、Ki-67和cyclin A的高表达及p27的低表达具有相同的生物学意义。
Objective To investigate the expression of cyclooxygenase-2 (COX-2) and cyclin A (p27) in esophageal squamous cell carcinoma (ESCC) and their correlation. Methods Totally 102 resected specimens of ESCC and 28 normal squamous epithelial tissues adjacent to cancer were selected. The expressions of COX-2, Ki-67, cyclin A and p27 were detected by immunohistochemistry. Results The expression of COX-2, Ki-67 and cyclin A in ESCC was significantly higher than that in normal squamous epithelium (P <0.01), while the expression of p27 in ESCC was significantly lower than that in normal squamous epithelium (P <0.01) . The expression of COX-2 was correlated with the T stage of tumor. The expression of COX-2 in stage T1 ~ T2 was lower than that in stage T3 ~ T4 (P <0.05). The expression of COX-2 was correlated with cell differentiation -2 was positively correlated with Ki-67 and cyclin A expression, but negatively correlated with p27 expression. Conclusions The expression of COX-2 can reflect the depth of tumor cell infiltration in ESCC. The high expression of COX-2, Ki-67 and cyclin A and the low expression of p27 have the same biological significance.
