目的:探讨血管紧张素Ⅱ(Ang Ⅱ)在肾间质纤维化过程中的作用机制。 方法:应用MTT 比色法检测Ang Ⅱ对成纤维细胞的增生,RTPCR 检测纤溶酶原激活物抑制物(PAI1)mRNA 的表达。 结果:Ang Ⅱ可促进肾间质成纤维细胞增生及PAI1 mRNA 的表达,且氯沙坦(10 -4g/L) 可抑制Ang Ⅱ的促增生作用。 结论:Ang Ⅱ可能通过直接作用于肾间质成纤维细胞,促进其增生,同时抑制细胞外基质的降解而参与肾间质纤维化的过程,而Ang Ⅱ受体拮抗剂拮抗Ang Ⅱ促细胞增生作用。
Objective: To investigate the mechanism of angiotensin Ⅱ (Ang Ⅱ) in renal interstitial fibrosis. Methods: The proliferation of fibroblasts was detected by MTT colorimetric assay, and the expression of plasminogen activator inhibitor (PAI-1) mRNA was detected by RT-PCR. RESULTS: Ang Ⅱ promoted the proliferation of renal interstitial fibroblasts and the expression of PAI-1 mRNA. Losartan (10 -4 g / L) could inhibit the growth-promoting effect of Ang Ⅱ. CONCLUSION: Ang Ⅱ may participate in the process of renal interstitial fibrosis by directly acting on interstitial fibroblasts, promoting its proliferation and inhibiting the degradation of extracellular matrix. However, Ang Ⅱ antagonist antagonizes Ang Ⅱ promoting cell proliferation effect.
