据文献报道,mCALPAIN(CALCIUM ACTIVATED NEUTRAL PROTEINASE Ⅱ)在轴突及髓鞘的演变中起关键作用,并与细胞骨骼蛋白质的更新和退变有关.为研究这种蛋白酶在脑缺血时神经元受损中所起的作用,用免疫细胞化学方法检查了这种酶在脑缺血后的表达.取10只SD大白鼠,按CHEN等氏改良法造成大脑单侧局部缺血动物模型(永久性阻断左侧大脑中动脉的主支的同时,暂时阻断双侧颈总动脉).动物分二批,存活中各为6/小时及24小时,行心内灌注固定.取大脑运动区按常规作恒冷箱冰冻切片,片厚40μm,每6片取1片,贴片按ABC法行免疫细胞化学法检验.另一套片用NISSL染色.在大脑运动区尼氏染色及mCALPAIN样免疫反应性均在局部缺血区与非缺血区出现明显分界线.6小时组在缺血区Ⅱ~Ⅴ层神经元的尼氏染色减弱,而mCALPAIN样免疫反应性明显增强,并随存活时间增加而更甚,24小时组的尼点染色更弱,而mCALPAIN样免疫反应性更强.结果提示,这种蛋白酶可能与迟发性神经元损害有关.与我们以前研究实验性脊髓损伤动物的结果相似.
