目的 通过观察氨茶碱对体外不同密度嗜酸性粒细胞 (Eos)凋亡及FasmRNA表达的影响 ,探讨其促进哮喘Eos凋亡的机制。方法 卵蛋白激发哮喘豚鼠动物模型 4 8h后行支气管肺泡灌洗 ,分离不同密度Eos。氨茶碱 (AM )( 10 -6 ~ 10 -3mol·L-1)干预 2 4h ,原位杂交检测Eos的FasmRNA表达 ,TUNEL法检测细胞凋亡。结果 AM干预 2 4h ,可见Eos凋亡增加 ,以低密度Eos(HEos)为明显 ,与不加药物相比 ,在 10 -5mol·L-1时差异有显著性 (P <0 0 5 ) ,FasmRNA表达降低 ,呈剂量依赖性 ,HEos与正常密度 (NEos)相比 ,其FasmRNA表达无差别。FasmRNA的表达与Eos凋亡呈正相关。结论 AM可提高FasmRNA表达 ,促进Eos凋亡。Fas可能是AM调节Eos凋亡的通路之一。
Objective To investigate the effect of aminophylline on the apoptosis of Eos and the expression of FasmRNA in vitro, and to explore its mechanism of promoting apoptosis of Eos in asthma. Methods The ovalbumin-induced asthmatic guinea pigs were treated with bronchoalveolar lavage for 48 hours and isolated Eos at different densities. Aminophylline (AM) (10 -6 ~ 10 -3 mol·L-1) for 24 h, the expression of Fas mRNA in Eos was detected by in situ hybridization and the apoptosis was detected by TUNEL method. RESULTS: After 24 h of AM intervention, the apoptosis of Eos increased, with the low density of Eos (HEos) being obvious. Compared with no drug, the difference was significant at 10 -5 mol·L-1 (P <0.05) Fas mRNA expression decreased in a dose-dependent manner. There was no difference in the expression of Fas mRNA between HEos and normal tissue (NEos). Fas mRNA expression was positively correlated with Eos apoptosis. Conclusion AM can increase the expression of Fas mRNA and promote the apoptosis of Eos. Fas may be one of the pathways by which AM regulates Eos apoptosis.
