建立了以羧甲基-β-环糊精(CM-β-CD)作为手性流动相添加剂,高效液相色谱拆分标题化合物的方法。系统考察了流动相中添加剂的种类及浓度、有机修饰剂、pH、三乙胺的含量和流速等因素对拆分的影响。色谱分离条件:流动相(V(甲醇)∶V(乙腈)∶V(水)=15∶70∶15),CM-β-CD浓度为0.6 g/L,三乙胺含量为0.14%(V/V),pH为3.92,流速为0.2mL/min,检测波长214 nm,药物对映体得到良好分离。
A method for the resolution of the title compound using high performance liquid chromatography with carboxymethyl-β-cyclodextrin (CM-β-CD) as a chiral mobile phase additive was established. The influences of the kind and concentration of additives in the mobile phase, organic modifiers, pH, the content of triethylamine and the flow rate on the resolution were systematically investigated. Chromatography conditions: mobile phase (V (methanol): V (acetonitrile): V (water) = 15:70:15), CM-β-CD concentration 0.6 g / L, triethylamine content 0.14% / V), the pH was 3.92, the flow rate was 0.2 mL / min, the detection wavelength was 214 nm, and the enantiomers of the drug were well separated.
