目的:建立同时测定夏枯草药材中咖啡酸、迷迭香酸、齐墩果酸及熊果酸含量的高效液相色谱方法。方法:夏枯草样品以75%乙醇溶液(含1%甲酸)超声提取,分析色谱柱为Elite SinoChrom ODS-AP柱(4.6 mm×250 mm,5μm),以0.01%磷酸溶液-乙腈为流动相进行梯度洗脱;采用波长切换方式检测,330 nm(0~33 min),203 nm(33~40 min);柱温20℃,进样量50μL。结果:咖啡酸、迷迭香酸、齐墩果酸与熊果酸的峰面积与其质量浓度之间均具有良好的线性关系,平均加样回收率93.7%~105.2%,RSD值均小于4.5%,所测夏枯草样品中咖啡酸、迷迭香酸、齐墩果酸与熊果酸的质量分数分别为0.040 1~0.096 8,0.99~2.57,0.243~0.556,4.06~8.13 mg.g-1。结论:本方法简便、快速、准确,可同时测定夏枯草中咖啡酸、迷迭香酸、齐墩果酸和熊果酸的含量。
Objective: To establish a HPLC method for simultaneous determination of caffeic acid, rosmarinic acid, oleanolic acid and ursolic acid in Prunella vulgaris. METHODS: Prunella vulgaris samples were extracted with 75% ethanol solution (containing 1% formic acid). The analytical column was Elite SinoChrom ODS-AP column (4.6 mm × 250 mm, 5 μm) with 0.01% phosphoric acid solution as mobile phase Gradient elution; detected by wavelength switching method, 330 nm (0 ~ 33 min), 203 nm (33 ~ 40 min); column temperature 20 ℃, injection volume 50μL. Results: There was a good linear relationship between the peak areas of caffeic acid, rosmarinic acid, oleanolic acid and ursolic acid and its mass concentration. The average recoveries were 93.7% -105.2%, RSD values were less than 4.5% . The measured contents of caffeic acid, rosmarinic acid, oleanolic acid and ursolic acid in the samples of Prunella vulgaris were 0.040 1 ~ 0.096 8, 0.99 ~ 2.57, 0.243 ~ 0.556, 4.06 ~ 8.13 mg.g -1 . Conclusion: The method is simple, rapid and accurate and can be used for the simultaneous determination of caffeic acid, rosmarinic acid, oleanolic acid and ursolic acid in Prunella vulgaris.
