目的:探讨膜联蛋白A2(ANXA2)在卵巢癌细胞中的表达及其功能。方法:常规培养卵巢癌细胞株SKOV3,取正常卵巢细胞为正常对照组,RT-PCR法检测不同细胞中ANXA2的表达,并采用MTT法检测两种细胞的存活率。结果:SK-OV3细胞株内ANXA2mRNA含量(89.13±3.66)%明显高于正常卵巢细胞(10.09±1.41)%(P<0.05),转染siRNA后,SK-OV3细胞株内ANXA2mRNA较转染前明显降低(21.13±2.54)%(P<0.05)。转染阴性siRNA SKOV3细胞株内的ANXA2mR-NA(88.21±3.54)%与未转染siRNA SKOV3细胞株内的ANXA2mRNA水平相当(P>0.05)。SKOV3细胞株存活率(70.71±5.34)%较正常卵巢细胞(55.35±4.71)%明显增高,转染siRNA后SKOV3细胞株存活率下降到(52.19±4.97)%与正常卵巢细胞的存活率相似(P>0.05),正常卵巢细胞转染siRNA后其存活率(49.93±5.67)%无明显改变(P>0.05)。结论:ANXA2的高表达参与卵巢癌的发生发展。
Objective: To investigate the expression and function of annexin A2 (ANXA2) in ovarian cancer cells. Methods: The ovarian cancer cell line SKOV3 was cultured routinely. The normal ovarian cells were taken as normal control group. The expression of ANXA2 in different cells was detected by RT-PCR. The survival rates of the two cell lines were detected by MTT assay. Results: The ANXA2 mRNA expression in SK-OV3 cells was significantly higher than that in normal ovary cells (89.13 ± 3.66% vs 10.09 ± 1.41%, P <0.05) (21.13 ± 2.54)% (P <0.05). ANXA2mR-NA (88.21 ± 3.54)% in transfected negative siRNA SKOV3 cells was comparable to that of ANXA2 mRNA in untransfected siRNA SKOV3 cells (P> 0.05). The survival rate of SKOV3 cell line (70.71 ± 5.34)% was significantly higher than that of normal ovary cells (55.35 ± 4.71)%, the survival rate of SKOV3 cell line decreased to (52.19 ± 4.97)% after transfected with siRNA was similar to that of normal ovarian cells P> 0.05). The survival rate of normal ovary cells transfected with siRNA (49.93 ± 5.67)% had no significant change (P> 0.05). Conclusion: The high expression of ANXA2 is involved in the development of ovarian cancer.
