骨癌痛大鼠髓内SIGIRR的表达变化

目的本研究针对目前日益增长的骨癌痛病患的疼痛问题进行试验探讨。建立癌痛动物模型寻找有效缓解疼痛的方法,进一步探讨骨癌痛大鼠模型中髓内细胞蛋白SIGIRR的表达水平的变化。方法建立SD大鼠胫骨癌痛模型,随机分为空白组(10只)和骨癌痛模型组(10只)。通过大鼠痛行为学测试、影像学、组织学方法对大鼠胫骨癌痛的模型进行验证。运用RT-PCR和Western blot检测髓内蛋白细胞LTR4和SIGIRR表达变化。结果 Walker256大鼠乳腺癌细胞制备大鼠胫骨癌痛的模型骨癌造模成功。模型组的体质量不增或者降低(P<0.01)、SAPS显著增加(P<0.01)、PWTL差异不大(P>0.05)。RT-PCR检测的TLR4结果显示,骨癌痛模型组比空白组表达明显增加。Western blot检测骨癌痛模型组比空白组髓内蛋白细胞SIGIRR明显下降。结论骨癌痛大鼠髓内蛋白细胞SIGIRR表达的降低可能与骨癌痛的发生机制相关。 Objective This study aimed to investigate the current pain problem of bone cancer pain patients. To establish an animal model of cancer pain to find effective ways to relieve pain and to further investigate the expression of intracellular marrow cell protein SIGIRR in rat model of bone cancer pain. Methods SD rat model of tibial cancer pain was established and randomly divided into blank group (n = 10) and bone cancer pain model group (n = 10). The rat model of tibial cancer pain was validated by the rat pain behavior test, imaging and histological methods. The expression of LTR4 and SIGIRR in intramedullary protein was detected by RT-PCR and Western blot. Results Walker256 rat breast cancer cell model of rat bone cancer pain in the success of bone cancer modeling. The body weight of the model group did not increase or decrease (P <0.01), SAPS increased significantly (P <0.01), PWTL difference was not significant (P> 0.05). The results of TLR4 assay by RT-PCR showed that the bone cancer pain model group than the blank group was significantly increased. Western blot detection of bone cancer pain model group than the blank group intramedullary protein cells SIGIRR decreased significantly. Conclusion The decrease of SIGIRR expression in intramedullary protein in bone cancer pain rats may be related to the mechanism of bone cancer pain.
