目的探讨银杏叶提取物对家兔动脉粥样硬化形成的影响以及作用机制。方法 23只新西兰大白兔随机分为对照组、假手术组、粥样硬化模型组(AS)、银杏叶提取物组(GBE)。采用高脂饮食联合球囊损伤复制家兔动脉粥样硬化模型,药物干预4周,抽血检测各组血脂水平;病理切片HE染色;免疫组织化学法和半定量逆转录-聚合酶链式反应(RT-PCR)法检测单核细胞趋化蛋白-1(MCP-1)蛋白和mRNA表达。结果银杏叶提取物(100mg/kg)对高脂血症无明显抑制作用(P>0.05),但可明显抑制斑块形成(P<0.05),下调MCP-1mRNA和蛋白表达(P<0.05)。结论银杏叶提取物对动脉粥样硬化病变有一定的抑制作用,这种作用可能与下调MCP-1的表达有关。
Objective To investigate the effect of Ginkgo biloba extract on atherosclerosis in rabbits and its mechanism. Methods Twenty - three New Zealand white rabbits were randomly divided into control group, sham operation group, atherosclerosis model group (GB) and ginkgo biloba extract group (GBE). High-fat diet and balloon injury were used to replicate atherosclerosis model in rabbits. Drugs were given for 4 weeks to detect blood lipid levels in each group. Pathological sections were stained with HE, immunohistochemistry and semi-quantitative reverse transcription-polymerase chain reaction (MCP-1) protein and mRNA were detected by RT-PCR. Results Ginkgo biloba extract (100 mg / kg) had no significant inhibitory effect on hyperlipidemia (P> 0.05), but inhibited the formation of plaque (P <0.05) and downregulated the expression of MCP-1 mRNA and protein . Conclusion Ginkgo biloba extract has certain inhibitory effect on atherosclerotic lesions, which may be related to the down-regulation of MCP-1 expression.
