对侧柏叶精油提取的两种方法(水蒸气蒸馏法和微波萃取法)进行了比较研究,并在单因素试验基础上,通过响应面法优化了微波萃取工艺,同时利用3D-响应曲面图及其等高线图,对影响微波萃取侧柏叶精油提取率的关键因素及其相互作用进行探讨,确定了微波萃取侧柏叶精油的最佳工艺。最后用气相色谱质谱联用仪(GC-MS)分别对传统的水蒸气蒸馏法和采用优化后的微波萃取法提取的精油进行化学成分的差异分析。结果表明:微波萃取最佳工艺条件为:功率为1 800 W、时间为30 min、液料比为20︰1(mL/g),此微波萃取工艺相对于传统的水蒸气蒸馏法的优点为有效缩短了萃取时间,大大提高了精油的提取得率(提高了2.72%),萃取出的化合物种类明显增多(多了8种)。
The two methods (steam distillation and microwave extraction) of extraction of essential oil from Oriental arborvitae were compared and studied. The microwave extraction process was optimized by response surface methodology on the basis of single factor experiment. At the same time, And its contour map, the key factors influencing the extraction rate of the essential oil from Oriental Arborvitae Boil and its interaction were discussed, and the best technology of extracting essential oil from Arborvitae by microwave was determined. At last, the difference of chemical composition between the traditional steam distillation method and the essential oil extracted by the optimized microwave extraction method was analyzed respectively by the gas chromatography-mass spectrometry (GC-MS). The results showed that the optimum conditions of microwave extraction were as follows: the power was 1 800 W, the time was 30 min and the liquid / solid ratio was 20︰1 (mL / g).
