扶正化瘀方影响肝脏胶原生成的拆方配伍研究

将扶正化瘀方拆分为扶正组、化瘀组、虫草组、丹参组及丹参加虫草组。制备各组合药血清,分离培养大鼠肝星状细胞,观察含药血清对培养星状细胞增殖活化、Ⅰ型胶原蛋白及其mRNA表达的影响。复制二甲基亚硝胺大鼠肝纤维化模型,以全方及拆方组治疗,观察模型大鼠肝脏羟脯氨酸(Hyp)含量与胶原沉积的变化。结果各组含药血清均可抑制星状细胞的增殖和活化,其中化瘀药效果最明显;各组含药血清均可抑制星状细胞Ⅰ型胶原mRNA及其蛋白的生成,以扶正组效果最佳。对DMN模型肝纤维化的治疗中,以全方组降低肝组织Hyp含量与胶原沉积最为明显,化瘀药与扶正药配伍后减轻胶原生成作用增强,全方具有较好的综合效果。说明扶正化瘀方有较好的抑制星状细胞增殖与胶原基因表达作用,是该方抗肝纤维化作用的重要机理,其中扶正药重在抑制胶原生成,化瘀药长于抑制星状细胞增殖,而全方具有综合效果。 Fuzheng Huayu Decoction was divided into Fuzheng group, Huayu group, Cordyceps group, Salvia miltiorrhiza group and Dan participated in Cordyceps group. Results The drug-containing serum of each group could inhibit the proliferation and activation of stellate cells, among which the effect of phlegm-resolving drugs was the most obvious. The serum containing drugs of each group could inhibit the production of type I collagen mRNA and protein in stellate cells, and the effect of the stiletto group was achieved. optimal. In the treatment of hepatic fibrosis with DMN model, the Hyp content and collagen deposition in hepatic tissue were the most obvious in the whole group, and the combination of Huayu medicine and Fuzheng medicine reduced the collagen production, and the whole formula had a better comprehensive effect. The results showed that Fuzheng Huayu Decoction had better inhibition of stellate cell proliferation and collagen gene expression, which was an important mechanism of the anti-hepatic fibrosis effect of the Fangzheng Huayu Decoction. Among them, Fuzheng Pills inhibited the production of collagen, and the pharmacokinetic drug was longer than the inhibition of stellate cell proliferation. , and all parties have a comprehensive effect.
