目的:研究生姜对局灶性脑缺血再灌注大鼠大脑皮层神经细胞凋亡和相关基因表达的影响。方法:SD雄性大鼠随机分为假手术组、模型对照组、尼莫地平组、生姜高、中、低剂量组。采用线栓法造成大鼠局灶性脑缺血再灌注模型(MCAO),分别于手术前24 h、术前1 h和再灌注后12 h灌胃给药,共3次。积分法测定MCAO大鼠神经病学症状,末端脱氧核糖核酸介导生物素化脱氧尿嘧啶缺口末端标记(TUNEL)法测定大脑皮层凋亡神经细胞,免疫组织化学方法研究大脑皮层神经细胞caspase-3,Bcl-2和Bax蛋白表达。结果:与假手术组比较,MCAO大鼠大脑皮层凋亡细胞明显增加,caspase-3和Bax蛋白表达增强,Bcl-2/Bax显著降低;生姜能明显降低MCAO大鼠皮层TUNELc,aspase,Bax和Bcl-2阳性细胞数,Bcl-2/Bax显著升高。结论:生姜可通过抑制神经细胞凋亡途径实现对脑缺血及缺血再灌注损伤的保护作用。
Objective: To study the effects of Ginger on neuronal apoptosis and related gene expression in cerebral cortex after focal cerebral ischemia / reperfusion in rats. Methods: SD male rats were randomly divided into sham operation group, model control group, nimodipine group, ginger high, medium and low dose groups. The focal cerebral ischemia-reperfusion model (MCAO) was induced by thread occlusion in rats. The rats were administered intragastrically for 24 h, 1 h before surgery and 12 h after reperfusion, respectively. Integral method was used to determine the neurological symptoms in MCAO rats. Neuronal apoptosis of cerebral cortex was detected by terminal deoxynucleotidyl transferase mediated dUTP nick end labeling (TUNEL). The expression of caspase-3, Bcl-2 and Bax protein expression. Results: Compared with the sham-operated group, the apoptotic cells in the cortex of MCAO rats were significantly increased, while the expressions of caspase-3 and Bax proteins were increased and Bcl-2 / Bax was significantly decreased. Ginger significantly reduced the levels of TUNELc, aspase, Bcl-2 positive cells, Bcl-2 / Bax increased significantly. Conclusion: Ginger can protect the cerebral ischemia and ischemia-reperfusion injury by inhibiting the apoptosis of nerve cells.
