目的:研究微型染色体维持蛋白2(MCM2)和Fas相关磷酸酯酶-1(FAP-1)在肝细胞肝癌组织中的表达及其与肝细胞肝癌增殖和凋亡的关系。方法:免疫组化SP法检测40例肝细胞肝癌、15例癌旁肝硬化和10例正常肝组织中MCM2和FAP-1蛋白的表达水平并进行分析。结果:40例肝细胞肝癌组织中29例MCM2表达阳性,28例FAP-1表达阳性,而在癌旁肝硬化和正常肝组织中两者均未见表达。肝细胞肝癌组织中MCM2的表达与患者性别、年龄、肿瘤大小和转移差异无统计学意义,P>0.05;而与分化程度有关,肿瘤分化越低,MCM2阳性率越高,P<0.05。MCM2与FAP-1两者呈正相关,P<0.05。结论:MCM2和FAP-1可能均参与了肝细胞肝癌的发生发展,两者可能相互协同共同促进了癌细胞异常增殖。
Objective: To investigate the expression of MCM2 and Fas-related phosphatase-1 (FAP-1) in hepatocellular carcinoma and its relationship with the proliferation and apoptosis of hepatocellular carcinoma. Methods: The expression of MCM2 and FAP-1 protein in 40 cases of hepatocellular carcinoma, 15 cases of paracancerous cirrhosis and 10 cases of normal liver tissues were detected by immunohistochemical SP method and analyzed. Results: The expression of MCM2 was positive in 29 cases of hepatocellular carcinoma and 28 cases of positive expression of FAP-1, but not in the adjacent cirrhotic and normal liver tissues. The expression of MCM2 in hepatocellular carcinoma was not related to the gender, age, tumor size and metastasis (P> 0.05), but correlated with the degree of differentiation. The lower the tumor differentiation was, the higher the MCM2 positive rate was (P <0.05). There was a positive correlation between MCM2 and FAP-1, P <0.05. Conclusion: Both MCM2 and FAP-1 may be involved in the development of hepatocellular carcinoma. Both of them may cooperate with each other to promote the abnormal proliferation of cancer cells.
