目的:研究清络通痹颗粒(QLT)对佐剂性关节炎大鼠骨质破坏的影响。方法:制备佐剂性关节炎(AIA)大鼠模型。自模型制备第7天开始给予不同剂量的QLT,持续49d。各组大鼠进行关节摄片,观察骨质破坏情况。取各组大鼠滑膜细胞培养上清液,分别加入体外培养的软骨细胞中,观察软骨细胞增殖反应、软骨细胞上清液中一氧化氮(NO)含量及诱导型一氧化氮合酶(iNOS)表达的变化情况。结果:QLT3个剂量组可不同程度地改善AIA大鼠后肢关节的骨密度降低、关节腔间隙增宽、关节结构破坏等表现;QLT7.2、14.4g.kg-1给药的AIA大鼠滑膜细胞培养上清液可增强软骨细胞的增殖能力,降低其iNOS的表达和NO的含量。结论:一定剂量的QLT对AIA大鼠的软骨和骨质破坏有抑制作用。
Objective: To study the effect of Qingluo Tongbi Granules (QLT) on bone destruction in adjuvant arthritis rats. Methods: A rat model of adjuvant arthritis (AIA) was prepared. Different doses of QLT were given from the 7th day of model preparation for 49 days. The rats in each group were photographed with joints to observe the bone destruction. The supernatants of rat synovial cell cultures were harvested and added to chondrocytes cultured in vitro. The chondrocyte proliferation reaction, nitric oxide (NO) content in chondrocyte supernatant, and inducible nitric oxide synthase were observed. iNOS) changes in expression. RESULTS: QLT 3 dose groups could improve bone mineral density, articular cavity widening, and joint structure destruction of hindlimb joints in AIA rats to varying degrees; ALA rats administered QLT7.2 and 14.4 g.kg-1 were slippery. Membrane cell culture supernatants can enhance the proliferation of chondrocytes and reduce their iNOS expression and NO content. Conclusion: A certain dose of QLT can inhibit the cartilage and bone destruction of AIA rats.
