通过不同病因诱导的肝纤维化模型,从蛋白、核酸两个方面观察基质金属蛋白酶Ⅱ(matrix metalloproteinase 2,MMP-2)表达状况,综合分析在不同病因作用下,MMP-2表达变化规律,探讨其在肝纤维化演进过程中的作用。一、材料与方法1.动物模型的建立和标本收集:Wistar雄性大鼠,(15±10)g,40只,超清洁级,复旦大学医学院实验动物中心提供。动物随机分成两组,每组20只,其中实验鼠15只,对照鼠5只。分别用四氯化碳和猪血清建立肝纤维化动物模型。
To observe the expression of matrix metalloproteinase 2 (MMP-2) from two aspects of protein and nucleic acid through different etiological models of liver fibrosis, and to analyze the changes of MMP-2 expression under different etiologies Its role in the evolution of liver fibrosis. Materials and Methods 1. Establishment of animal models and collection of specimens: Wistar male rats, (15 ± 10) g, 40, super clean grade, provided by Fudan University School of Medicine Laboratory Animal Center. Animals were randomly divided into two groups of 20 rats each, including 15 experimental mice and 5 control mice. Animal models of liver fibrosis were established with carbon tetrachloride and porcine serum, respectively.
