目的:研究中药注射液血必净对感染性休克犬抗凝物质的影响。方法:通过给健康杂种犬静脉内注射内毒素制造感染性休克的动物模型,以收缩压下降较注射内毒素前大于40mmHg(1mmHg=0.133 3kPa)开始计时,持续稳定60min,即视为模型成功。按照随机原则将制模成功的动物分为3组:多巴胺治疗组6只,血必净+多巴胺治疗组(简称血必净组)6只,另设对照组6只。分别按照实验设计的时间点即注射内毒素前(基础),模型成功后用药前,以及用药后3,7h检测抗凝血酶Ⅲ(antithrombinⅢ,AT-Ⅲ)、组织纤溶酶原激活物(Tissue-type plasminogen activator,TPA)和血栓调节蛋白(thrombomodu-line,TM)血浆含量指标,采用SPSS统计软件进行统计分析。结果:犬机体抗凝功能监测指标AT-Ⅲ、TPA以及TM:血必净组与对照组、多巴胺组相比,在用药后3或7h3项监测指标变化差异具有显著性(P<0.01或P<0.05),增加了这些物质的血浆浓度;多巴胺组只有TM在3h与对照组相比差异具有显著性(P<0.05)。结论:在应用多巴胺治疗感染性休克犬的基础上加用血必净可有效的增加感染性休克犬体内的抗凝物质血浆水平而稳定凝血系统,可能是中药注射液血必净对感染性休克犬发挥治疗作用的机制之一。
Objective: To study the effect of Xuebijing injection on anticoagulant substances in septic shock dogs. Methods: Animal models of septic shock were made by intravenous injection of endotoxin into healthy mongrel dogs. The systolic blood pressure was decreased by more than 40 mmHg (1 mmHg = 0.133 3 kPa) before the injection of endotoxin, and continued to be stable for 60 minutes. According to the principle of randomness, the successful animals were divided into three groups: 6 in the dopamine group, 6 in the Xuebijing + dopamine group (Xuebijing group) and 6 in the control group. The effects of antithrombin Ⅲ (AT-Ⅲ), tissue plasminogen activator Tissue-type plasminogen activator (TPA) and thrombomodu-line (TM) plasma were measured by SPSS statistical software. Results: The monitoring indicators of anticoagulant function of AT-Ⅲ, TPA and TM: Xuebijing group were significantly different from those of the control group and the dopamine group (P <0.01 or P <0.05), and increased plasma concentrations of these substances. Only the TM in the dopamine group at 3h was significantly different from the control group (P <0.05). Conclusion: The application of Xuebijing in the treatment of septic shock dogs with dopamine can effectively increase the plasma level of anticoagulant substance in septic shock dogs and stabilize the coagulation system, which may be due to the effect of Xuebijing Injection on septic shock Dogs play one of the mechanisms of treatment.
