目的:以人白血病细胞系HL60和小鼠B16BL6黑色素瘤细胞株为对象,以细胞抑制率、细胞凋亡为指标,评价白花蛇舌草提取物及多糖的体外诱导癌细胞凋亡效应,为白花蛇舌草的抗肿瘤效应提供实验依据。方法:采用水提取物、乙醇提取物及多糖提取物,应用MTT和XTT法检测提取物对B16BL6、HL60肿瘤细胞株的细胞抑制率,采用FCM法检测其诱导细胞凋亡的效果。结果:(1)MIT/XTT法定量检测,3种提取物对肿瘤均有一定的抑制作用,且抑制作用随药物浓度的增加而增大;(2)FCM法检测,白花蛇舌草醇提物和多糖对B16BL6细胞凋亡和细胞周期的影响与空白组比较差异有显著性。结论:白花蛇舌草水提物、醇提物和多糖对小鼠B16BL6黑色素瘤细胞和HL60细胞株均具有明显的抑制作用,醇提物和多糖具有明显的诱导细胞凋亡作用。
OBJECTIVE: To evaluate the apoptosis-inducing effect of Hedyotis diffusa Willd. Extract and polysaccharide in vitro on human leukemia cell line HL60 and mouse B16BL6 melanoma cell line by using cell inhibition and apoptosis as indexes, The anti-tumor effect of Hedyotis diffusa provides experimental evidence. Methods: The water extract, ethanol extract and polysaccharide extract were used to detect the inhibitory rate of the extracts on the B16BL6 and HL60 tumor cell lines by MTT and XTT methods. The effect of the extract on the apoptosis was detected by FCM. Results: (1) MIT / XTT assay showed that the three extracts had certain inhibitory effect on the tumor and the inhibitory effect increased with the increase of the drug concentration. (2) FCM assay, Hedyotis diffusa alcohol extract The effects of polysaccharide and polysaccharide on apoptosis and cell cycle of B16BL6 cells were significantly different from those of blank group. Conclusion: The aqueous extract of Hedyotis diffusa will, ethanol extract and polysaccharide have obvious inhibitory effect on the B16BL6 melanoma cells and HL60 cell lines, alcohol extract and polysaccharides have obvious induction of apoptosis.
