建立了高效液相色谱-串联质谱(HPLC-MS/MS)检测肉牛毛发中氯丙那林残留的方法。牛毛发经1 mol/L盐酸溶液水解、MCX固相萃取柱净化,用HPLC-MS/MS进行检测。氯丙那林在0.2~800 ng/m L浓度范围内线性关系良好(r≥0.999);氯丙那林的回收范围为95.9%~99.1%,日内变异系数范围为1.1%~7.7%,日间变异系数范围为5.5%~12.1%,检测限为0.25μg/kg,定量限为1.0μg/kg。应用该方法初步研究和探讨了氯丙那林在牛毛发中的代谢规律,结果表明喂药5 d至停药21 d牛毛发中均检出氯丙那林,氯丙那林容易在毛发中蓄积,且代谢缓慢,毛发可作为肉牛饲养过程中对氯丙那林药物监管的有效靶组织。
A high performance liquid chromatography-tandem mass spectrometry (HPLC-MS / MS) method for the determination of clorpranarin residues in beef cattle hair was established. The bovine hair was hydrolyzed by 1 mol / L hydrochloric acid solution and cleaned up by MCX solid phase extraction column, and detected by HPLC-MS / MS. The linear range of chlorpromazine was 0.2 ~ 800 ng / m L (r≥0.999), the recovery range of chlorpromazine was 95.9% ~ 99.1%, and the coefficient of variation (CV) ranged from 1.1% to 7.7% The coefficient of variation (CV) ranged from 5.5% to 12.1% with a detection limit of 0.25 μg / kg and a limit of quantification of 1.0 μg / kg. The method was used to preliminary study and discuss the metabolism of chlorpromazine in cow’s hair. The results showed that clindrenaline was detected in 5 days after feeding and 21 days after withdrawal. Chlorpromazine was easily found in the hair Accumulation, and slow metabolism, hair can be used as beef cattle feeding regimen of chlorpromazine effective target tissue.
