目的:探讨5-FU结合大黄虫汤诱导胃癌细胞凋亡的作用。方法:将人胃癌BGC—823细胞裸鼠移植瘤动物模型,随机分为4组(每组12只):5-FU结合大黄虫汤组,大黄虫汤组,5-FU组,生理盐水组。连续用药3周,用药结束后脱颈处死裸鼠,统计各组抑瘤率;流式细胞仪检测移植瘤细胞凋亡率,RT-PCR检测凋亡关键因子Caspase-3表达。结果:5-FU结合大黄虫汤组抑瘤率高于大黄虫汤组、5-FU组(P<0.01);5-FU结合大黄虫汤组胃癌细胞凋亡率明显高于大黄虫汤组、5-FU组(P<0.01,P<0.05),5-FU结合大黄虫汤组Caspase-3水平最高,与大黄虫汤组、5-FU组比较有显著差异(P<0.01,P<0.05)。结论:5-FU结合大黄虫汤能提高胃癌细胞凋亡率,抑制胃癌的增长。
Objective: To investigate the role of 5-FU in combination with Rhubarb and Oviduct in inducing gastric cancer cell apoptosis. Methods: The animal model of BGC-823 human gastric cancer xenografts in nude mice was randomly divided into 4 groups (12 in each group): 5-FU combined with Rhubarb, Saline group. The mice were sacrificed 3 weeks after the treatment. The nude mice were sacrificed and the tumor inhibition rate was calculated. The rate of apoptotic cells was detected by flow cytometry and the expression of Caspase-3, a key apoptosis factor, was detected by RT-PCR. Results: The inhibition rates of 5-FU combined with Rhubarb and Oviduct soup were higher than that of Rhubarb and Zhifu Tang, 5-FU group (P <0.01). The apoptosis rate of gastric cancer cells was significantly higher than that of Rhubarb (P <0.01, P <0.05). Caspase-3 level was the highest in 5-FU combined with rhubarb-emperor wormwood soup group, which was significantly different from those in Dahuangzhitang decoction group and 5-FU group P <0.01, P <0.05). Conclusion: 5-FU combined with Rhubarb and Oviduct can increase the apoptosis rate of gastric cancer cells and inhibit the growth of gastric cancer.
