目的建立绝卵巢切除术后皮肤老化模型,观测黄芪提取液对卵巢切除术后大鼠皮肤衰老的影响。方法将60只Wister雌性大鼠随机分成4组:非手术组、手术组(切除双侧卵巢正常喂养组)、雌激素给药组和黄芪给药组。正常饮食、饮水,饲育50d后处死大鼠,取皮肤组织测定其生物活性酶及病理组织观察。结果与非手术组比较,手术组皮肤组织中SOD活性明显降低,羟脯氨酸含量显著减少,丙二醛含量显著增高(P<0.05)。雌激素组及黄芪组皮肤组织中SOD活性不降低,羟脯氨酸含量无减少,丙二醛含量不增高(P>0.05)。与手术组比较,黄芪组皮肤组织中SOD活性明显增高,羟脯氨酸含量增加,丙二醛含量降低(P<0.05),与雌激素组无显著性差异(P>0.05)。组织病理示:,卵巢切除术后皮肤组织老化,雌激素和黄芪治疗后可改善和延缓其皮肤衰老。结论切除大鼠双侧卵巢可以建立绝经期皮肤老化模型,黄芪提取液对大鼠皮肤衰老具有延缓作用,与激素替代疗法无显著性差异。
Objective To establish a model of skin aging after ovariectomy and observe the effects of astragalus extract on skin aging in ovariectomized rats. Methods Sixty female Wister rats were randomly divided into 4 groups: non-operation group, operation group (normal ovariectomized group), estrogen administration group and astragalus administration group. Normal diet, water, rats were sacrificed 50d after feeding, take the skin tissue to determine its biological activity of enzymes and pathological observation. Results Compared with the nonsurgical group, the activity of SOD in the skin tissue of the operation group was significantly decreased, the content of hydroxyproline was significantly decreased and the content of malondialdehyde was significantly increased (P <0.05). The activity of SOD in the estrogen group and the astragalus group did not decrease, the content of hydroxyproline did not decrease and the content of malondialdehyde did not increase (P> 0.05). Compared with the operation group, the activity of SOD in the skin of Astragalus membranaceus group was significantly increased, the content of hydroxyproline increased, the content of malondialdehyde decreased (P <0.05), but there was no significant difference with the estrogen group (P> 0.05). Histopathology showed: skin aging after ovariectomy, estrogen and astragalus treatment can improve and delay the aging of the skin. Conclusion Ablation of bilateral ovaries can establish a model of menopausal skin aging. Astragalus extract has a retardation effect on skin aging in rats, which is not significantly different from hormone replacement therapy.
