用荧光光谱和紫外可见吸收光谱法研究了在不同温度下,中草药黄芩的水提取液中的活性成分(RS)与牛血清白蛋白(BSA)的相互作用的光谱行为,试验发现,RS对BSA有较强的荧光猝灭作用。用Stern-Volmer和Lineweaver-Burk方程分别处理试验数据,发现RS与BSA反应生成新的复合物且发生了分子内的非辐射能量转移,属于是静态荧光猝灭类型,并计算出不同温度下形成复合物的形成常数KLB(288 K:4.538×10~4 L·mol~(-1);308 K:3.488×10~4L·mol~(-1))及对应温度下结合反应的热力学参数(△H~θ=-9.71 kJ·mol~(-1);△S~θ=55.45/55.42kJ·mol~(-1);△G~θ=25.68/-26.78 kJ·mol~(-1)),证明二者主要靠疏水作用力结合。这为了解中草药中的活性小分子与蛋白质等生物大分子的弱相互作用提供有益的参考和依据。
The spectroscopic behavior of the interaction between active ingredient (RS) and bovine serum albumin (BSA) in the aqueous extract of Scutellaria baicalensis Georgi at different temperatures was studied by fluorescence spectroscopy and UV-Vis absorption spectroscopy.
