目的:研究中药注射剂的过敏反应,测定致敏动物血清IgE抗体和组织胺含量,观察其与过敏反应是否具有相关性,为提高I型过敏反应试验预测的准确性提供实验依据。方法:选用3种中药注射剂进行PCA,ASA试验和类过敏反应研究,用ELISA法对致敏动物血清进行血清OVA-sIgE、血清总IgE、组织胺水平检测。结果:3种中药注射剂的PCA试验结果为阴性,血清总IgE水平与生理盐水组无明显差异;3种中药注射剂+OVA(不引起过敏反应的剂量)的研究中双黄连注射液(SHL)、鱼腥草注射液(YXC)的PCA试验均为阳性,OVA-sIgE水平明显增加,清开灵注射液(QKL)的PCA试验为阴性,OVA-sIgE水平增加不明显,各组血清总IgE水平增加不明显;3种中药注射剂的ASA试验结果为阳性;3种中药注射剂(临床剂量和临床等效剂量)均引起了豚鼠的类过敏症状,且反应强弱呈现剂量和注射速度依赖性,各给药组血清组织胺水平均较生理盐水组明显增高,血清总IgE水平无明显差异。结论:SHL和YXC具有增加豚鼠对卵蛋白敏感性的作用,SHL,YXC,QKL均能引起豚鼠的类过敏反应,3种中药注射剂的过敏反应的发生与特异性IgE抗体和组织胺水平有相关性。
OBJECTIVE: To study the anaphylactic reaction of traditional Chinese medicine (TCM) injection and to determine the serum IgE and histamine contents in sensitized animals and to observe whether it has correlation with allergic reactions and to provide experimental evidence for improving the prediction accuracy of type I allergic reactions. Methods: Three kinds of traditional Chinese medicine injections were used to study PCA, ASA and anaphylactoid reactions. Serum levels of OVA-sIgE, total serum IgE and histamine were detected by ELISA. Results: The PCA test results of three kinds of traditional Chinese medicine injections were negative, and there was no significant difference between the two groups in the total serum IgE level and the saline group. Shuanghuanglian injection (SHL), three kinds of traditional Chinese medicine injection + OVA (dose not causing allergic reaction) The PCA test of Yuxingcao injection (YXC) was positive, the level of OVA-sIgE was significantly increased, the PCA test of Qingkailing injection (QKL) was negative and the level of OVA-sIgE did not increase significantly. The levels of serum total IgE ASA test results of three kinds of traditional Chinese medicine injections were positive. Three kinds of traditional Chinese medicine injections (clinical dose and clinical equivalent dose) all caused the allergic symptoms in guinea pigs, and the response rate was dose-dependent and injection-rate-dependent The level of serum histamine in the administration group was significantly higher than that in the normal saline group, and there was no significant difference in the total serum IgE level. CONCLUSIONS: SHL and YXC can increase the sensitivity of guinea pigs to ovalbumin. SHL, YXC and QKL can cause allergic reactions in guinea pigs. The incidence of allergic reactions to three Chinese traditional medicine injection is related to specific IgE and histamine levels Sex.
