目的:观察光照改变对雄性金黄地鼠下丘脑-垂体-性腺轴的影响。方法:2日龄雄性金黄地鼠40只,随机分为正常组(12h,50lux)、16h弱光组(16h,50lux)、16h强光组(16h,350lux)和20h强光组(20h,350lux),光照干预至28日龄;放免法检测血清睾酮(T)、黄体生成素(LH)、促卵泡激素(FSH)水平;Real-time PCR法检测下丘脑促性腺激素释放激素(Gn RH)、促性腺激素抑制激素(Gn IH)、褪黑素受体1c亚型(Mel1c),垂体Gn RHR、GPR147 m RNA表达变化。结果:光照组金黄地鼠出现躁动不安,皮毛光泽度差,体质量偏轻等类似阴虚火旺证候表现。与正常组比较,各光照组T水平显著上升(P<0.05);下丘脑Mel1c m RNA表达水平显著下降(P<0.01,P<0.05);16h弱光组和20h强光组下丘脑Gn IH m RNA表达水平显著升高(P<0.05)。结论:延长光照时间、增加光照强度可使雄性地鼠出现一定的阴虚火旺表现。光照时间延长、强度增加可引起雄性地鼠睾丸发育提前,其机制可能与褪黑素受体、Gn IH有关。
Objective: To observe the effects of light changes on the hypothalamus-pituitary-gonad axis of male hamster. Methods Forty two-day-old male hamsters were randomly divided into normal group (12h, 50lux), 16h low light group (16h, 50lux), 16h light group (16h 350lux) and 20h light group 350 lux) were given to the rats for 28 days. The levels of serum testosterone (LH) and follicle stimulating hormone (FSH) were measured by radioimmunoassay. The levels of gonadotropin-releasing hormone ), Gn IH, Mel1c, pituitary Gn RHR and GPR147 m RNA expression. Results: The golden hamsters in the light group showed restlessness, poor glossiness of the fur, and mild body weight. Compared with the normal group, the T level in each light group increased significantly (P <0.05), the expression level of Mel1c m RNA in hypothalamus decreased significantly (P <0.01, P <0.05) m RNA expression was significantly increased (P <0.05). Conclusion: To prolong the illumination time and increase the light intensity, the male rats showed certain Yin deficiency and Huo wang performance. The lengthening of illumination time and the increase of intensity can lead to the development of testis in male hamster, which may be related to the melatonin receptor and Gn IH.
