目的:研究红花黄色素对神经根型腰椎病大鼠抗炎作用及其机制。方法:建立神经根型腰椎病大鼠模型,随机分为假手术组、模型组、PDTC(吡咯烷二硫代甲酸铵盐)组、红花黄色素100 mg/kg、50 mg/kg、25 mg/kg剂量组。红花黄色素各组于造模后3 d静脉注射给药,共给药7 d。检测后肢热痛刺激缩爪阈值的变化;椎板法检测血液流变学指标;Elisa法检测大鼠血清中TNF-α、IL-1β、IL-6、COX-2水平;Western法检测脊髓背角中p-NF-k B P65蛋白的表达;RT-PCR法检测NF-k B P65 mRNA的表达。结果:与模型组相比,100 mg/kg、50 mg/kg、25 mg/kg剂量的红花黄色素均能显著提高热痛刺激缩爪阈值、改善血液流变学各指标;显著降低TNF-α、IL-1β、IL-6、COX-2水平;显著抑制p-NF-k B P65蛋白及NF-k B P65 mRNA的表达。结论:红花黄色素可能对神经根型腰椎病的治疗有一定的意义,其机制与抑制NF-k B P65通路的激活有关。
Objective: To study the anti-inflammatory effect and its mechanism of safflower yellow on rat with lumbar nerve root radiculopathy. Methods: A rat model of lumbar radiculopathy was established and randomly divided into sham-operation group, model group, PDTC (pyrrolidine dithiocarbamate) group, safflower yellow pigment 100 mg / kg, 50 mg / kg, 25 mg / kg dose group. Each group of safflower yellow veins were injected intravenously 3 days after modeling for a total of 7 days. Serum levels of TNF-α, IL-1β, IL-6 and COX-2 were measured by Elisa method; Western blot was used to detect spinal cord dorsal index The expression of p-NF-κB P65 protein was detected by RT-PCR. The expression of NF-κB P65 mRNA was detected by RT-PCR. Results: Compared with the model group, the safflower yellow dye of 100 mg / kg, 50 mg / kg and 25 mg / kg significantly increased the threshold value of thermal paw withdrawal stimulation, and improved the indexes of hemorrheology. -α, IL-1β, IL-6 and COX-2, and significantly inhibited the expression of p-NF-κB P65 protein and NF-κB P65 mRNA. Conclusion: Safflower yellow pigment may have some significance for the treatment of radiculopathy of radiculopathy, and its mechanism is related to the inhibition of the activation of NF-κB P65 pathway.
