目的建立苦参药材中黄酮类成分的HPLC指纹图谱方法,为科学全面评价该药材质量提供依据。方法采用HPLC法对不同产地的苦参药材进行分析。色谱条件为Alltech Apollo C18色谱柱(4.6 mm×250 mm,5μm),流动相为甲醇-0.2%甲酸水溶液系统,梯度洗脱,流速1.0 mL/min,检测波长310 nm,柱温25℃。结果确定了13个色谱峰(或峰组)为各产地药材特征共有峰,并利用对照品对照和LC-MS法对其中的6个共有峰进行了成分指认。结论该方法简便可靠,重复性好,能够表征苦参药材的内在信息,可用于苦参药材中黄酮类成分的质量控制。
OBJECTIVE To establish HPLC fingerprinting method of Flavonoids in Sophora flavescens, and to provide a scientific basis for comprehensively evaluating its quality. Methods HPLC method was used to analyze Sophora flavescens from different areas. The chromatographic conditions were Alltech Apollo C18 column (4.6 mm × 250 mm, 5 μm). The mobile phase consisted of methanol-0.2% formic acid aqueous solution with gradient elution at a flow rate of 1.0 mL / min. The detection wavelength was set at 310 nm and the column temperature was 25 ℃. As a result, 13 peaks (or peak groups) were identified as the common peaks of the medicinal materials in each place of production, and the 6 common peaks among them were identified by reference substance control and LC-MS method. Conclusion The method is simple, reliable and reproducible. It can characterize the intrinsic information of kushen herbs and can be used for the quality control of flavonoids in kushen herbs.
