目的:探讨尿路上皮癌胚抗原Ⅰ(UCA1)mRNA在舌鳞状细胞癌中的表达及其与舌鳞癌病理分级和临床分期的关系。方法:采用SYBR Green II实时荧光定量反转录聚合酶链反应实验方法,从6种基因中筛选出在40对舌鳞癌组织及配对正常舌组织样本中表达有明显差异的基因UCA1,扩大样本量至93例,检测UCA1 mRNA在两者中的表达。应用SPSS13.0软件包分析UCA1 mRNA表达量与舌鳞癌患者临床病理学特征的关系。结果:UCA1 mRNA在93例患者的肿瘤组织中平均表达量为0.7213,在正常舌组织中为0.2756,表达量有显著差异(P<0.001);UCA1 mRNA的表达与患者的性别、年龄、肿瘤大小无关(P>0.05),与舌鳞癌的病理分级和临床分期有显著相关性(P<0.05)。结论:UCA1 mRNA高表达与舌鳞状细胞癌密切相关,对舌鳞癌的诊断、判断病理分级和临床分期具有重要意义。
Objective: To investigate the expression of UCA1 mRNA in tongue squamous cell carcinoma and its relationship with pathological grade and clinical stage of tongue squamous cell carcinoma. Methods: SYBR Green II real-time quantitative reverse transcription-polymerase chain reaction assay was used to screen out the gene UCA1 in 40 pairs of tongue squamous cell carcinoma tissues and paired normal tongue tissues, and to amplify the sample To 93 cases, the detection of UCA1 mRNA expression in both. The relationship between the expression of UCA1 mRNA and clinicopathological characteristics of tongue squamous cell carcinoma was analyzed by SPSS 13.0 software package. Results: The average expression level of UCA1 mRNA in tumor tissue of 93 patients was 0.7213, and in normal tongue tissue was 0.2756, the expression level was significantly different (P <0.001). The expression of UCA1 mRNA was positively correlated with the gender, age, tumor size (P> 0.05), and it was significantly correlated with the pathological grade and clinical stage of tongue squamous cell carcinoma (P <0.05). Conclusion: The high expression of UCA1 mRNA is closely related to tongue squamous cell carcinoma, which is of great significance for the diagnosis of tongue squamous cell carcinoma, pathological grade and clinical stage.
