目的:探讨夏枯草提取物对T淋巴瘤细胞模型小鼠抑瘤作用及其对T淋巴瘤EL-4细胞生长的影响。方法:优选BALB/C小鼠30只为研究对象,通过腹腔注射EL-4瘤株建立T淋巴瘤细胞小鼠模型。随机将其分为健康对照组、低剂量夏枯草、中剂量夏枯草、高剂量夏枯草、环磷酸胺组以及T淋巴瘤模型组,每组小鼠各5只。分别应用TUNEL法、MTT法、DNA琼脂糖凝胶电泳法分析夏枯草提取物对模型小鼠抑瘤率、细胞凋亡情况及细胞凋亡率的影响。结果:(1)体重分析:夏枯草中、高剂量组用药后小鼠体重与正常对照组无统计学意义(P>0.05),环磷酸胺组小鼠与对照组相比体重显著下降,差异有统计学意义(P<0.05)。(2)抑瘤率:夏枯草提取物中、高剂量组抑瘤率高于低剂量组及环磷酸胺组,差异有统计学意义(P<0.05)。(3)TUNEL染色面积分析:夏枯草低剂量组及环磷酸胺组TUNEL染色中多见细胞核呈黄色浓染区,而中高剂量组及空白对照组仅出现个别棕黄色染色,且低剂量组及环磷酸胺组阳性区分光密度、阳性区面积、阳性平均灰度量化高于中高剂量组及空白对照组(P<0.05)。(4)Bax蛋白及BC2蛋白表达:夏枯草提取物中高剂量组与环磷酸胺组相比,Bax蛋白表达增加,而BC2蛋白表达上升,差异有统计学意义(P<0.05)。结论:夏枯草提取物能有效抑制T淋巴瘤细胞生长,促进细胞肿瘤细胞凋亡,其作用机制可能与其调控Bax蛋白及BC2蛋白表达有关。
Objective: To investigate the anti-tumor effect of Prunella vulgaris extract on T lymphoma model mice and its effect on the growth of T lymphoma EL-4 cells. Methods: Totally 30 BALB / C mice were selected as the study object. T lymphoma mouse models were established by intraperitoneal injection of EL-4 tumor cells. Randomly divided into healthy control group, low dose of Prunella vulgaris, middle dose of Prunella vulgaris, high dose of Prunella vulgaris, cyclophosphamide group and T lymphoma model group, 5 mice in each group. TUNEL method, MTT method and DNA agarose gel electrophoresis were used to analyze the effect of Prunella vulgaris extract on tumor inhibition rate, apoptosis and apoptosis rate in model mice. Body weight analysis: There was no significant difference between the body weight of the middle and high dose Prunella vulgaris groups and the normal control group (P> 0.05), and the body weight of the cyclophosphamide group was significantly lower than that of the control group There was statistical significance (P <0.05). (2) The inhibition rate of tumor: Prunella extract, high dose group of tumor inhibition rate was higher than low dose group and cyclophosphamide group, the difference was statistically significant (P <0.05). (3) TUNEL staining area analysis: Prunella low dose group and cyclophosphamide group TUNEL staining showed yellow staining area, while the middle and high dose group and the blank control group, only a few brown color, and low dose group and Cyclophosphamide positive group differentiated optical density, positive area, positive mean gray value was higher than the high dose group and the blank control group (P <0.05). (4) Bax protein and BC2 protein expression: Compared with cyclophosphamide group, the expression of Bax protein and BC2 protein in Prunella vulgaris extract medium and high dose group increased significantly, the difference was statistically significant (P <0.05). Conclusion: Prunella vulgaris extract can effectively inhibit the growth of T lymphoma cells and promote the apoptosis of tumor cells. The mechanism may be related to the regulation of Bax protein and BC2 protein expression.
