目的探讨脐带间充质干细胞(UC-MSCs)体外对白血病、淋巴瘤、多发性骨髓瘤8种血液肿瘤细胞株增殖的影响。方法用羧基荧光素二乙酸盐琥珀酰亚胺酯(CFDA-SE)标记肿瘤细胞,UC-MSCs与肿瘤细胞以不同比例体外共培养作为实验组,以肿瘤细胞单独培养作为对照组,流式细胞仪检测3 d后肿瘤细胞的平均荧光强度及分裂的代次。结果 UC-MSCs对HL60、THP1、K562、RPMI8226细胞增殖起抑制作用;对Raji及NCIH929细胞增殖起促进作用;对Jurkat细胞仅在1∶1时起促进作用;而对于U937细胞,在UC-MSCs与U937比例为2∶1时起抑制作用,1∶4及1∶16时起促进作用,1∶1时无明显影响。结论UC-MSCs对不同血液肿瘤细胞株增殖的作用不同。对4株白血病细胞系无明显的促进作用,对淋巴瘤细胞株及多发性骨髓瘤细胞株具有促进和抑制的双向作用,U937细胞株可能是进一步研究这种对立现象的良好模型。
Objective To investigate the effects of umbilical cord mesenchymal stem cells (UC-MSCs) on the proliferation of 8 hematological malignant tumor cell lines with leukemia, lymphoma and multiple myeloma in vitro. Methods The tumor cells were labeled with carboxyfluorescein diacetate succinimidyl ester (CFDA-SE). UC-MSCs and tumor cells were co-cultured with different ratios in vitro as experimental group and tumor cells cultured alone as control group. Flow cytometry The average fluorescence intensity of tumor cells and the generation of division after 3 days were detected by cytometry. Results UC-MSCs could inhibit the proliferation of HL60, THP1, K562 and RPMI8226 cells, promote the proliferation of Raji and NCIH929 cells, and promote the proliferation of Jurkat cells only at 1: 1. For U937 cells, With U937 ratio of 2: 1 from the inhibition, 1: 4 and 1:16 from the role of promotion, 1: 1 no significant effect. Conclusion UC-MSCs have different effects on the proliferation of different hematological tumor cell lines. On the four leukemia cell lines did not significantly promote the lymphoma cell lines and multiple myeloma cell lines have a two-way role in the promotion and inhibition, U937 cell lines may be further study of this opposite phenomenon of a good model.
