目的比较慈姑多糖和慈姑水提部位在体外对异烟肼(INH)和利福平(RFP)合用致肝细胞损伤的保护作用。方法采用系统化学溶剂萃取法制备慈姑水提部位,采用水提醇沉法制备慈姑多糖。培养Hep G2肝细胞,建立异烟肼和利福平合用(INH/RFP)致体外肝细胞损伤模型,给予慈姑水提部位或慈姑多糖处理后,利用MTT法检测肝细胞的存活率,并检测细胞培养上清液中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、乳酸脱氢酶(LDH)、丙二醛(MDA)的浓度。结果与模型组比较,慈姑水提部位组和慈姑多糖组均可增加细胞存活率(P<0.05),降低INH/RFP损伤肝细胞培养上清液中ALT、AST、LDH、MDA水平(P<0.05),慈姑多糖组较慈姑水提物部位组细胞存活率更高,细胞培养上清液中ALT、AST、LDH、MDA水平更低。结论慈姑多糖和慈姑水提部位对体外INH/RFP致肝细胞损伤均有保护作用,慈姑多糖作用更强,其作用可能与抗脂质过氧化有关。
Objective To compare the protective effects of aqueous extract from Sagittaria and Sagittaria on hepatocyte injury induced by isoniazid (INH) and rifampin (RFP) in vitro. Methods The aqueous extract of Sagittaria was prepared by systematic chemical solvent extraction method. Hep G2 hepatocytes were cultured and established in vitro hepatocyte injury model induced by isoniazid combined with rifampicin (INH / RFP). After treated with Arrowhead water extract or arrowhead polysaccharide, the survival rate of hepatocytes was detected by MTT assay Concentrations of alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH) and malondialdehyde (MDA) in cell culture supernatants. Results Compared with the model group, the cell viability and the ALT, AST, LDH and MDA levels in hepatocyte culture supernatants of INH / RFP group were both increased (P <0.05) 0.05). The cell viability of the Sagittaria polysaccharide group was higher than that of the Sagittaria aqueous extract group, and the levels of ALT, AST, LDH and MDA in the cell culture supernatant were lower. Conclusions The extracts from Sagittaria polyglucosae and Sagittaria giganteus have a protective effect on INH / RFP induced hepatocyte injury in vitro, and the effect of Sagittaria polysaccharides is stronger than that of Sagittaria polysaccharides, which may be related to the anti-lipid peroxidation.
