目的:运用均匀设计并结合定量指纹图技术优化丹黄凝胶醇提工艺参数。方法:采用HPLC指纹图谱技术,按U8(82×42)均匀设计考察乙醇体积分数、乙醇用量、提取次数和提取时间4个因素,以大黄酚和小檗碱含量、HPLC指纹图谱特征峰总面积之和以及干浸膏得率为综合评价指标,采用多元非线性拟合,建立效应指标和考察因素之间的数学模型,根据最佳数学模型描绘效应面图和等高线图,确定醇提工艺参数的优化范围及最佳工艺条件。结果:建立的数学模型较好的描述效应指标与因素之间的关系,优化的最佳醇提工艺条件为7倍量65%乙醇,提取3次,每次2 h。结论:运用定量指纹图技术并结合均匀设计和效应面分析优选的醇提工艺合理可行,为丹黄凝胶进一步研究提供了参考。
OBJECTIVE: To optimize the technical parameters of Dan Gelin by uniform design and quantitative fingerprinting.
