目的观察H1N1型流感病毒诱导的MDCK细胞抗氧化相关修复基因MTH1表达量的变化。方法 MDCK细胞培养后,H1N1型流感病毒感染0、1、3、6、12、24、48h,在各时间点经RT-PCR反应检测细胞内MTH1基因的相对表达量。结果甲型H1N1流感病毒感染后0、6、12h,MTH1基因的表达量与正常对照组差异无统计学意义;感染后1、3hMTH1基因的表达量显著高于正常对照组;感染后24、48h表达量显著低于正常对照组。结论 H1N1流感病毒感染细胞的MTH1基因表达的增加,可能增加细胞对DNA氧化损伤的修复能力,在流感发生和防御机制中起到作用。
Objective To observe the changes of MTH1 gene expression in MDCK cells induced by H1N1 influenza virus. Methods After MDCK cell culture, the H1N1 influenza virus was infected for 0, 1, 3, 6, 12, 24 and 48 hours. The relative expression level of MTH1 gene was detected by RT-PCR at each time point. Results The expression of MTH1 gene at 0, 6 and 12 h after influenza A (H1N1) infection was not significantly different from that of the normal control group. The mRNA expression of MTH1 at 1 and 3 h after infection was significantly higher than that of the control group. At 24 h and 48 h The expression level was significantly lower than the normal control group. Conclusion The increase of MTH1 gene expression in infected cells of H1N1 influenza virus may increase the ability of cells to repair DNA oxidative damage and play an important role in the pathogenesis and prevention of influenza.
