目的观察急性白血病骨髓基质细胞上connexin43蛋白的表达以及GJIC功能。方法体外培养正常及急性白血病骨髓基质细胞,采用激光共聚焦扫瞄显微镜观察二者Cx43表达的变化;采用荧光漂白恢复技术比较二者之间GJIC功能的差异。结果正常及急性白血病骨髓基质细胞上Cx43表达的像素密度分别为(81.04±8.84)%和(34.10±17.91)%,两组间差异有统计学意义(P<0.01)。结论急性白血病骨髓基质细胞间通讯功能较正常骨髓基质细胞明显减弱。
Objective To investigate the expression of connexin43 protein and the function of GJIC on acute leukemia bone marrow stromal cells. Methods Normal and acute leukemia bone marrow stromal cells were cultured in vitro. The changes of Cx43 expression were observed by laser scanning confocal microscopy. The differences of GJIC function between the two groups were compared using fluorescence bleaching recovery technique. Results The density of Cx43 expression in normal and acute leukemia bone marrow stromal cells was (81.04 ± 8.84)% and (34.10 ± 17.91)%, respectively. There was a significant difference between the two groups (P <0.01). Conclusion Acute leukemia bone marrow stromal cells compared with normal bone marrow stromal cells was significantly weakened.
