目的:探讨膜转运蛋白MRP1、LRP和BCRPmRNA在非小细胞肺癌(NSCLC)中的表达及临床预后意义。方法:用RT-PCR法检测60例NSCLC患者癌组织及癌旁组织中MRP1、LRP和BCRPmRNA的表达。用t检验、方差分析、秩相关分析、Kaplan-Meier生存曲线和COX多因素回归分析进行统计分析。结果:MRP1和LRPmRNA在NSCLC中的表达量高于癌旁组织(P=0.02和P=0.000),癌组织中两者表达呈正相关(rs=0.530,P=0.000),BCRPmRNA在癌组织中表达量低于癌旁组织(P=0.042)。不同组织类型中,肺腺癌LRPmRNA表达量高于鳞癌(P=0.02);不同临床分期癌组织中MRP1、LRP和BCRPmRNA表达均无差异。预后分析显示MRP1mRNA低表达组患者生存期明显长于高表达组(P=0.023),MRP1mRNA高表达是影响NSCLC患者预后的独立因素。结论:MRP1、LRP和BCRPmRNA在NSCLC组织中有不同程度的表达。MRP1mRNA表达水平与患者生存期相关,可作为判断NSCLC患者预后的一项指标。
Objective: To investigate the expression of MRP1, LRP and BCRP mRNA in non-small cell lung cancer (NSCLC) and their clinical significance. Methods: The expressions of MRP1, LRP and BCRP mRNA in 60 cases of NSCLC and their adjacent tissues were detected by RT-PCR. T-test, ANOVA, rank correlation analysis, Kaplan-Meier survival curve and COX multivariate regression analysis were used for statistical analysis. Results: The expression of MRP1 and LRP mRNA in NSCLC was significantly higher than that in paracancer tissues (P = 0.02 and P = 0.000). The expression of MRP1 and LRP mRNA in NSCLC was positively correlated (rs = 0.530, P = 0.000) Lower than the number of paracancerous tissues (P = 0.042). The expression of LRP mRNA in lung adenocarcinoma was higher than that in squamous cell carcinoma (P = 0.02) in different tissue types. There was no difference in MRP1, LRP and BCRP mRNA expression in different clinical stages. Prognostic analysis showed that MRP1mRNA expression in patients with low expression group was significantly longer than the high expression group (P = 0.023), MRP1mRNA expression is an independent factor affecting the prognosis of NSCLC patients. Conclusion: MRP1, LRP and BCRP mRNA are expressed in different degrees in NSCLC tissues. MRP1mRNA expression level is related to patient’s survival, which can be used as an index to judge the prognosis of NSCLC patients.
