目的研究溶血磷脂酸(LPA)对原代培养人支气管肺上皮细胞(NHBE)表达纤溶酶原激活物抑制因子-1(PAI-1)的调节作用。方法原代培养人支气管上皮细胞培养于6孔板培养板内,经LPA诱导后,收取RNA裂解液并用实时荧光定量RT-PCR技术检测PAI-1 mRNA表达水平;利用RNA干扰技术研究LPA受体的作用。结果 LPA可诱导NHBE细胞PAI-1 mRNA表达上调且呈时间相关性,此效应可被LPA2 siRNA完全阻断。结论 LPA2受体介导LPA诱导NHBE细胞表达PAI-1。
Objective To investigate the regulatory effect of lysophosphatidic acid (LPA) on expression of plasminogen activator inhibitor type 1 (PAI-1) in primary cultured human bronchial lung epithelial cells (NHBE). Methods Primary cultured human bronchial epithelial cells were cultured in 6-well plate. After induced by LPA, RNA lysate was collected and the expression of PAI-1 mRNA was detected by real-time fluorescence quantitative RT-PCR. LPA receptor Role. Results LPA could up-regulate PAI-1 mRNA expression in NHBE cells in a time-dependent manner. This effect was completely blocked by LPA2 siRNA. Conclusion LPA2 receptor mediates LPA-induced PAI-1 expression in NHBE cells.
