目的观察固本活血壮骨颗粒对糖尿病大鼠模型成骨细胞Wnt/β-catenin信号传导通路的影响。方法 6月龄SD大鼠随机分为正常对照组、模型对照组、阳性药物对照组、固本活血壮骨颗粒大、中、小剂量组。除正常对照组外,其余各组均采用左下腹腔内注射新鲜配制1%链脲佐菌素STZ(柠檬酸缓冲液,p H值4.4)65 mg/kg体重造模。各组予相应药物及生理盐水灌胃12周。结果固本活血壮骨颗粒可提高糖尿病模型成骨细胞Wnt1、LRP-5以及β-catenin蛋白的表达。结论固本活血壮骨颗粒可以通过促进成骨细胞Wnt1、LRP-5以及β-catenin蛋白的表达,调节Wnt/β-catenin信号传导通路的失调状态,促进骨形成,减少骨吸收,提高骨质量,从而达到防治糖尿病性骨质疏松症的效果。
Objective To observe the effects of Gubenhuoxuezhuanggu granules on the Wnt / β-catenin signaling pathway of osteoblasts in diabetic rat model. Methods Six-month-old SD rats were randomly divided into normal control group, model control group, positive drug control group, Gubenhuoxuezhuanggu Granule large, medium and small dose groups. Except for the normal control group, all the other groups were given a fresh preparation of 1% streptozotocin STZ (citrate buffer, p H value 4.4) at a dosage of 65 mg / kg body weight in the lower left abdominal cavity. The corresponding groups of drugs and saline for 12 weeks. Results Gubenhuoxuezhuanggu granules can increase the expression of Wnt1, LRP-5 and β-catenin in diabetic model. Conclusion Gubenhuoxuezhuanggu granule can regulate the imbalance of Wnt / β-catenin signaling pathway by promoting the expression of Wnt1, LRP-5 and β-catenin in osteoblasts, promote bone formation, reduce bone resorption and increase bone mass , So as to achieve the effect of preventing and treating diabetic osteoporosis.
