小白鼠是遗传学和药物毒理研究常用的动物之一。我们用小鼠骨髓直接制备染色体,不需要复杂和严格的培养条件,此方法简单、快速、分散程度良好,染色体清晰。操作方法介绍如下: 一、主要试剂: 1.2%柠檬酸钠。 2.秋水仙素:配成0.05%秋水仙素溶液,冰箱保存。 3.低渗液:0.3%氯化钾溶液。 4.固定液:1:3的冰醋酸和甲醇,临用现配。 5:磷酸盐缓冲液。 6.姬姆萨染液。二、方法: 1.用无菌注射器,在小鼠腹腔注入0.05%秋水仙素,按体重3~4毫克/公斤。 2.注入秋水仙素3~4小时后,拉颈椎致死。
