目的探讨胶质瘤干细胞生存微环境—壁龛的结构和分布特征。方法采用免疫组化法检测26例胶质瘤标本中CD133、Nestin、ABCG2阳性细胞及壁龛的表达,计算其阳性细胞及壁龛数,并分级,分析其与病理分级的相关性。结果低级别(Ⅱ级)12例,高级别14例(Ⅲ级10例、Ⅳ级4例)。CD133、Nestin、ABCG2阳性细胞及其壁龛多半分布于血管周围以及血管相对富集的肿瘤边缘,三者均与病理分级呈正相关(P<0.05或P<0.01)。结论 CD133、Nestin、ABCG2阳性细胞及壁龛均以血管周围分布为主,与病理分级呈正相关关系;胶质瘤干细胞壁龛在胶质瘤发生、发展和浸润扩散中起着重要作用。
Objective To investigate the structure and distribution of glioma stem cell niche - niche. Methods The expressions of CD133, Nestin, ABCG2 positive cells and niches in 26 glioma specimens were detected by immunohistochemistry. The numbers of positive cells and niches were calculated and classified into categories, and the correlations with pathological grade were analyzed. Results Low grade (grade Ⅱ) in 12 cases, high grade in 14 cases (grade Ⅲ in 10 cases, grade Ⅳ in 4 cases). CD133, Nestin, ABCG2 positive cells and their niches were mostly located in the periphery of the blood vessels and the edge of the tumors that were relatively enriched in blood vessels, all of which were positively correlated with pathological grade (P <0.05 or P <0.01). Conclusion CD133, Nestin, ABCG2 positive cells and niches are mainly distributed around the blood vessels, and positively correlated with the pathological grade. The niche of glioma stem cells plays an important role in the occurrence, development and infiltration of glioma.
