对不同地理来源的向日葵黑茎病菌的ITS基因、Actin基因和EF基因进行测序,结合GenBank中的同源序列,对这3种基因的序列进行比对分析和系统进化分析,探讨ITS、Actin以及EF基因用于向日葵黑茎病菌的种内鉴定以及其系统进化的可行性。结果表明:ITS序列和Actin序列比较保守,无法作为种内鉴定的依据。EF基因序列其变异几率比较高,不同地理来源的菌株在EF-1α基因序列上表现出了比较大的差异,可以用于种内亲缘关系的研究。
The ITS gene, Actin gene and EF gene were sequenced from different geographical origin of the sunflower black stem strain, and the sequences of these three genes were analyzed by phylogenetic analysis based on the homologous sequences in GenBank. ITS, Actin and EF gene for identification of sunflower black stem inoculum and its phylogenetic feasibility. The results showed that the sequences of ITS and Actin were conservative and could not be used as the basis for intraspecific identification. EF gene sequence has a high probability of mutation, and strains from different geographical origins have relatively large differences in the sequence of EF-1α gene and can be used to study the genetic relationship within the species.
