血清中的奎尼丁经自制硅胶萃取柱提纯后,直接点于硅胶G板上,以环己烷:氯仿:二乙胺(5:4:2)展开,冰醋酸蒸汽熏后,全尼丁于UV254灯下显天蓝色荧光斑点,刮取斑点于10ml离心管中,0.1NH2SO4定容、旋摇、离心,取上清液于Ex355nm、EM445nm测荧光强度。当奎尼丁点样量在0.1~8ug之间,线性关系良好(r=0.9998),血清中奎尼丁固相提取平均回收率为95.25±4.55%,RSD=4.78%。用此法测定了家兔的药-时曲线及药动学参数。
Quinidine in serum was purified by self-made silica gel column, directly on silica gel G plate, developed with cyclohexane: chloroform: diethylamine (5: 4: 2) Under UV254 light blue sky fluorescent spots, scraping spots in 10ml centrifuge tube, 0.1NH2SO4 constant volume, shake, centrifuged, the supernatant was measured at Ex355nm, EM445nm fluorescence intensity measured. The average recoveries of quinidine in serum were 95.25 ± 4.55% with RSD = 4 when the amount of quinidine was in the range of 0.1-8ug and the linearity was good (r = 0.9998) .78%. This method was used to determine the rabbit’s drug-time curve and pharmacokinetic parameters.
