目的 研究pcDNA3.1·PTH治疗甲状旁腺功能低下症时PTH的变化及PTH与pcDNA3.1·PTH剂量的关系。方法 手术去掉家兔甲状旁腺 ,建立甲状旁腺功能低下症模型。将已构建的人甲状旁腺激素基因pcD NA3.1·PTH以 10 0 μg/ml、30 0 μg/ml、5 0 0 μg/ml剂量注射模型家兔的骨骼肌内 ,测定注射后不同时间的血液PTH。结果 注射pcDNA3.1·PTH后 2 4h模型家兔的PTH升高 ,7d达高峰水平 ,并持续 2个月 ,以 30 0 μg/ml和 5 0 0 μg/ml为佳。结论 人甲状旁腺激素基因治疗甲状旁腺功能低下是有效的 ,且以 30 0 μg/ml和 5 0 0 μg/ml为最佳剂量
Objective To study the changes of PTH and the relationship between PTH and pcDNA3.1 · PTH dosage when pcDNA3.1 · PTH is used to treat hypoparathyroidism.
