目的 研究 p73基因在结肠癌中的改变及其意义 .方法 采用 PCR- SSCP方法检测 40例结肠癌及其癌旁非癌组织中 p73基因突变 ,采用定量 RT- PCR方法检测 p73基因的表达 .结果 1PCR- SSCP检测出 7例存在 p73等位基因缺失 ,未见突变 ;2定量 RT- PCR检测出 33例结肠癌呈高表达 ,7例呈中等表达 ;非癌组织中 2 6例呈低水平表达 ,14例呈中等水平表达 ;p73基因在结肠癌与癌旁组织中的表达存在显著性差异 (P<0 .0 1) .结论 p73基因以等位基因缺失、高表达方式参与结肠癌发生发展
Objective To study the alteration and significance of p73 gene in colon cancer. Methods PCR-SSCP method was used to detect p73 gene mutations in 40 cases of colon cancer and its adjacent noncancerous tissues. The expression of p73 gene was detected by quantitative RT-PCR. 1 PCR-SSCP detected 7 cases of deletion of p73 allele, no mutations; 2 quantitative RT-PCR detected 33 cases of high expression of colon cancer, 7 cases showed moderate expression; 26 cases of non-cancerous tissue showed low expression The expression of p73 gene was significantly different in colon cancer and para-carcinoma tissues (P<0.01). Conclusion p73 gene is involved in the development of colon cancer with allele deletion and high expression.
