目的 探讨慢性 B-淋巴细胞性白血病 (B chronic lymphatic leukemia,B- CL L)原癌基因MDM2 (murine double minute 2 )表达与 12号染色体三体 (+12 )畸变的关系。方法 采用流式免疫荧光技术对 2 0例慢性 B-淋巴细胞性白血病 MDM2蛋白表达进行定量分析 ,采用荧光原位杂交检测 +12畸变 ,并将结果与传统细胞遗传学检查进行比较。结果 在 6例病例中发现 +12的克隆 ,而且该 6例病例 MDM2蛋白表达量较其余 14例有显著增高 (P<0 .0 1)。结论 +12畸变可作为 B- CL L分型的独立指标 ,MDM2扩增与 +12畸变密切相关 ,可能是 B- CL L的发生、发展机理之一。
Objective To investigate the relationship between the expression of murine double minute 2 (CDM2) and the trisomy (12) in chronic B-lymphoblastic leukemia (B-CL L). Methods The expression of MDM2 protein in 20 cases of chronic B-lymphocytic leukemia was quantitatively analyzed by flow cytometry, and the +12 strain was detected by fluorescence in situ hybridization. The results were compared with the traditional cytogenetic tests. Results A total of +12 clones were found in 6 cases, and MDM2 protein expression was significantly higher in the 6 cases than in the remaining 14 cases (P <0.01). Conclusion +12 aberration can be used as an independent marker of B-CL L classification. MDM2 amplification is closely related to +12 distortion, which may be one of the mechanisms of the development of B-CL L.
