[目的] 观察软肝抗纤方对肝贮脂细胞(HSC T6)Ⅰ型胶原(Col -Ⅰ)mRNA表达的影响。[方法] 以软肝抗纤方灌胃大鼠制备的不同浓度含药血清,作用于HSC -T6 细胞48 h,应用逆转录定量PCR方法测定其对HSC T6细胞Col- I mRNA表达的影响。[结果] 软肝抗纤方不同浓度的含药血清(5%、10%、20%)与氯化钠组(0 .9%)比较,对HSC、T6细胞Col I mRNA表达,各组差异均有统计学意义,其中5%,10%组可抑制HSC T6细胞Col- ⅠmRNA表达水平。[结论] 软肝抗纤方可抑制HSC -T6细胞Col- I mRNA表达水平,具有抗肝纤维化作用。
[Objective] To observe the effect of Ruangan Kangxian Recipe on the expression of type I collagen (Col-I) mRNA in hepatic lipid-storage cells (HSC T6). [Methods] The serum containing different concentrations of Ruangan Kangxianfang was prepared and treated with HSC-T6 cells for 48 h. The effect of the serum on the expression of Col- I mRNA in HSC T6 cells was determined by reverse transcription quantitative PCR. [Results] The differences of the expression of Col I mRNA in HSC and T6 cells were compared between the groups with different concentrations of Ruangan Kangxian Recipe (5%, 10%, 20%) and sodium chloride group (0.9%). All were statistically significant, of which 5% and 10% could inhibit the expression of Col-I mRNA in HSC T6 cells. [Conclusion] Ruangan Anti-Fibrosis Decoction can inhibit the expression of Col- I mRNA in HSC-T6 cells and has anti-fibrotic effect.
