采用丙酮分部沉淀 ,三氯乙酸 (TCA)去蛋白 ,再经 Sephadex G- 75和 CM- Sephadex C- 50柱层析 ,从钝顶螺旋藻中分离纯化得到一种多糖化合物 SPPC- 1。经高压液相色谱和毛细管电泳鉴定为均一性组分。凝胶过滤法测得其相对分子质量为 7.4× 10 4 。糖组成分析表明其主要由鼠李糖组成 ,还含有少量的木糖、甘露糖和葡萄糖 ,摩尔比为 Rha:Xyl:Man:Glc=6 .2 4 :0 .37:0 .13:0 .71。元素分析表明 ,SPPC- 1含碳 4 2 .59% ,含氢 8.0 7% ,红外光谱分析表明 ,SPPC- 1含 α-型糖苷连接键。ESR研究表明 SPPA- 1具有清除 O-.2 自由基活性
The polysaccharide compound SPPC-1 was isolated and purified from Spirulina platensis by using acetone fraction precipitation and trichloroacetic acid (TCA) deproteinization, followed by Sephadex G-75 and CM-Sephadex C-50 column chromatography. High pressure liquid chromatography and capillary electrophoresis identified as a homogeneous component. The relative molecular mass was 7.4 × 10 4 measured by gel filtration. Sugar composition analysis showed that it mainly consists of rhamnose, and also contains a small amount of xylose, mannose and glucose, the molar ratio of Rha: Xyl: Man: Glc = 6.24: 0.37: 0.13: 0. 71. Elemental analysis showed that SPPC-1 contained 42.259% of carbon and 8.07% of hydrogen, and IR spectra showed that SPPC-1 contained α-type glycosidic linkages. ESR studies have shown that SPPA-1 has a scavenging O-.2 free radical activity
