目的:探讨肿瘤坏死因子α(TNF-α)基因-238G>A及-857C>T多态性与河南汉族原发性高血压(EH)的相关性.方法:ELISA法检测260例高血压患者和394例健康对照者血浆TNF-α水平,采用限制性扩增片段长度多态性(PCR-RFLP)的方法检测TNF-α基因-238G>A及-857C>T多态性.结果:与正常对照组相比,高血压患者血浆TNF-α水平显著增高(P<0.01);EH组中TNF-α-238G>AGA+AA基因型者的DBP、TC、TG、及LDL含量明显高于GG纯合子者,差异有统计学意义(P<0.05),但是EH组TNF-α基因-238G>A及-857C>T各基因型及等位基因频率与正常对照组比较差异无统计学意义(P>0.05).结论:TNF-α-238G>AA等位基因可能会增加高血压的发病风险,但TNF-α基因-238G>A及-857C>T基因多态性与河南汉族原发性高血压无关联.
Objective: To investigate the correlation between the polymorphisms of -238G> A and -857C> T of tumor necrosis factor-α (TNF-α) gene and essential hypertension (EH) in Han nationality in Henan Province.METHODS: 260 cases of hypertensive patients TNF-α and TNF-α levels in 394 healthy controls were detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) .238G> A and -857C> Compared with normal control group, plasma TNF-αlevels in hypertensive patients were significantly increased (P <0.01); DBP, TC, TG, and LDL levels in TNF-α-238G> AGA + AA genotypes in EH group were significantly higher than those in normal controls GG homozygotes, the difference was statistically significant (P <0.05), but EH group of TNF-α gene -238G> A and -857C> T genotypes and allele frequencies compared with the normal control group, no significant difference (P> 0.05) .Conclusion: TNF-α-238G> AA allele may increase the risk of hypertension, but the polymorphisms of -238G> A and -857C> T genes of TNF- Hypertension no correlation.
