目的建立同时测定浙江蜡梅中3种黄酮类成分含量的高效液相色谱法。方法采用Agilent Zorbax SB-C18色谱柱(4.6 mm×250 mm,5μm),柱温为30℃;流动相为乙腈-0.1%磷酸溶液,梯度洗脱,流速为1.0 m L·min-1;检测波长为360 nm。结果芦丁、槲皮素和山奈酚的线性范围分别为4.057~507.2,0.686 1~171.5,0.216 9~81.35μg·m L-1,r分别为0.999 3,0.999 6,0.999 9;平均加样回收率分别为99.97%(RSD=0.97%),100.13%(RSD=1.06%),99.35%(RSD=1.39%)。结论该方法同时测定浙江蜡梅中的芦丁、槲皮素和山奈酚的含量,简便易行、快速准确、重复性好,可用于浙江蜡梅的质量控制。
Objective To establish a HPLC method for the simultaneous determination of three flavonoids in Chimonanthus praecox. Methods The Agilent Zorbax SB-C18 column (4.6 mm × 250 mm, 5 μm) was used and the column temperature was 30 ℃. The mobile phase consisted of acetonitrile-0.1% phosphoric acid solution with gradient elution at a flow rate of 1.0 mL · min-1. The wavelength is 360 nm. Results The linear ranges of rutin, quercetin and kaempferol were 4.057 ~ 507.2,0.686 1 ~ 171.5,0.216 9 ~ 81.35μg · m L-1, r were 0.999 3,0.999 6,0.999 9 respectively; The recoveries were 99.97% (RSD = 0.97%), 100.13% (RSD = 1.06%) and 99.35% (RSD = 1.39%), respectively. Conclusions The method for simultaneous determination of rutin, quercetin and kaempferol in Chimonanthus praecox is simple, rapid, accurate, reproducible and can be used for the quality control of Chimonanthus praecox.
