目的:建立用高效液相色谱法梯度洗脱同时测定脉络宁注射液中绿原酸、新绿原酸、隐绿原酸、咖啡酸、肉桂酸、3,4-二-O-咖啡酰基奎宁酸,3,5-二-O-咖啡酰基奎宁酸,4,5-二-O-咖啡酰基奎宁酸含量的方法,并对3个批次脉络宁注射液中上述8种化合物进行定量分析。方法:流动相乙腈-0.8%醋酸,梯度洗脱,流速1 mL·min-1,柱温30℃,检测波长λ1=327 nm(绿原酸、新绿原酸、隐绿原酸、咖啡酸、3,4-二-O-咖啡酰基奎宁酸,3,5-二-O-咖啡酰基奎宁酸,4,5-二-O-咖啡酰基奎宁酸),λ2=278 nm(肉桂酸);进样量为10μL。结果:绿原酸、新绿原酸、隐绿原酸、咖啡酸、肉桂酸、3,4-二-O-咖啡酰基奎宁酸,3,5-二-O-咖啡酰基奎宁酸,4,5-二-O-咖啡酰基奎宁酸的线性范围分别为2.57~25.7(r=0.999 7),2.05~20.5(r=0.999 7),2.26~22.6(r=0.999 9),1.92~19.2(r=0.999 9),1.16~11.6(r=0.999 7),2.282~22.82(r=0.999 8),1.839~18.39(r=0.999 8),2.194~21.94(r=0.999 7)mg·L-1,加样回收率分别为95.26%(RSD 1.21%),96.03%(RSD 1.09%),97.25%(RSD 2.18%),100.98%(RSD 1.98%),97.44%(RSD 2.38%),97.78%(RSD 2.24%),98.37%(RSD 2.15%),95.82%(RSD2.30%)。结论:方法简便、快捷,准确、重复性好,可为脉络宁注射液提供质量控制依据。
OBJECTIVE: To establish a method for the simultaneous determination of chlorogenic acid, neogenic acid, cryptocholic acid, caffeic acid, cinnamic acid, 3,4-di-O-caffeoyl quinine in Mailuoning injection by gradient elution Acid, 3,5-di-O-caffeoylquinic acid and 4,5-di-O-caffeoylquinic acid, and the above 8 compounds in 3 batches of Mailuoning injection were quantified analysis. METHODS: The mobile phase consisted of acetonitrile-0.8% acetic acid with gradient elution at a flow rate of 1 mL · min-1 and the column temperature was 30 ℃. The detection wavelength was λ1 = 327 nm (chlorogenic acid, neogenic chlorogenic acid, cryptoceric acid, caffeic acid, Di-O-caffeoylquinic acid, 3,5-di-O-caffeoylquinic acid, 4,5-di-O-caffeoylquinic acid), λ2 = 278 nm ); Injection volume of 10μL. Results: Chlorogenic acid, neogenic acid, cryptocholic acid, caffeic acid, cinnamic acid, 3,4-di-O-caffeoylquinic acid, 3,5-di-O-caffeoylquinic acid, 4 (R = 0.999 7), 2.05 ~ 20.5 (r = 0.999 7), 2.26 ~ 22.6 (r = 0.999 9), 1.92 ~ 19.2 (r = 0.999 9), 1.16-11.6 (r = 0.999 7), 2.282-22.82 (r = 0.999 8), 1.839-18.39 (r = 0.999 8), 2.194-21.94 (r = 0.999 7) mg · L- 1, the recoveries were 95.26% (RSD 1.21%), 96.03% (RSD 1.09%), 97.25% (RSD 2.18%), 100.98% (RSD 1.98%), 97.44% (RSD 2.38%), 97.78% (RSD 2.24%), 98.37% (RSD 2.15%), 95.82% (RSD 2.30%). Conclusion: The method is simple, rapid, accurate, reproducible and can provide basis for quality control of Mailuoning injection.
