目的:观察鼻渊舒口服液对兔CRS模型血清IL-8、TNF-α含量的影响,初步探索鼻渊舒口服液治疗CRS的可能机制。方法:取新西兰大白兔30只,适应性喂养1周后,随机分为正常组、模型组、假手术组、鼻渊舒组、西药治疗组,每组6只,建立CRS模型;鼻渊舒组、西药组分别给予鼻渊舒口服液1.5mL/(kg.d),克拉霉素25mg/(kg.d),共14天,处死后取鼻窦黏膜组织HE染色光镜观察,取血清ELISA法检测IL-8、TNF-α含量。结果:模型组鼻黏膜呈慢性炎症表现,黏膜上皮细胞增生,炎细胞浸润、腺体和杯状细胞明显增生;血清IL-8、TNF-α较正常组显著增高(P<0.05)。经鼻渊舒治疗后鼻黏膜上皮修复较好,炎细胞浸润、腺体和杯状细胞增生不明显;血清IL-8、TNF-α较模型组显著降低(P<0.05),与正常组比较无显著差异(P>0.05)。结论:鼻渊舒口服液对CRS具有确切的治疗作用,其机制与减轻IL-8、TNF-α导致的过度炎症反应有关。
OBJECTIVE: To observe the effect of Biyuan Shu oral solution on serum IL-8 and TNF-α levels in CRS model of rabbits and to explore the possible mechanism of Bi Yuan Shu oral solution in treating CRS. Methods: Thirty New Zealand white rabbits were randomly divided into three groups: normal group, model group, sham operation group, Biyuan Shu group and western medicine group. Six rabbits in each group were given CRS model. Group and western medicine group were given nasopharyngeal oral solution 1.5mL / (kg.d), clarithromycin 25mg / (kg.d), a total of 14 days, after sacrifice sinus mucosa HE staining, serum ELISA Method to detect IL-8, TNF-α content. Results: The nasal mucosa of the model group showed chronic inflammation, proliferation of epithelial cells, infiltration of inflammatory cells and glandular cells and goblet cells. Serum levels of IL-8 and TNF-α were significantly higher than those of the normal group (P <0.05). The nasal mucosa epithelium was repaired well after nasal and deep soothing treatment, inflammatory cell infiltration and glandular and goblet cell proliferation were not obvious. Serum IL-8 and TNF-α levels were significantly lower than those in model group (P <0.05) No significant difference (P> 0.05). CONCLUSION: Biyuan Shu oral solution has the exact therapeutic effect on CRS, and its mechanism is related to the reduction of excessive inflammatory reaction caused by IL-8 and TNF-α.
